At which cell density do you seed them out? Sometimes, this phenomenon occurs, when cells, especially adherent cells, are seeded out with a too low density.
Dogan is right, it is probably because your seeding density is too low. L6 cells do not migrate, they just divide, so if the seeding density is too low, you will just have few colonies apart from each other.
When I seed L6 cells, I prepare a suspension of cells around 200,000 cells/mL and add 1mL in 12-well plates or 2 mL in 6-well plates. Cells are usually confluent within the next two days. Try playing with different seeding densities and find what's the most convenient for you.
