I am planing to use a kit [(from cell biolabs (cat # STA-347) ] for the analysis of total ROS. In this kit it is given to sonicate or homogenize the cells but we don't have sonicator in our lab. So, please suggest me how to homogenize it or any other alternative for the lysis..
Although this will not help you now, you can buy the diacetate versions of the reagent and then measure ROS in living cells. This removes the need to lyse. As for lysing your cells, are they mammalian, bacterial, yeast?
Try use syringe with 21 gauge needle to shear the cells. This may not be perfect but works for small number of cells after syringe up-and-down 7-10 times.
The kit instructions lists a bunch of compatible detergents/denaturants. Triton-x works well for lysing cells.

Cell lysis is mainly depending upon integrity of cell. For mammalian cell , low detergent concentrations are sufficient to cause cell lysis (e.g., 1% Nonidet P-40 [NP-40] or 1% Triton X-100), this method can be gentler to the protein of interest than mechanical homogenization methods.
