Hi, I want to some miRNA study like miRNA profiling and microarray studies for So, I have to extract miRNA from cells like macrophage. please suggest a method with reference for that. Thank you..
The most commonly used method is probably TRIzol (Ambion) however a column-based kit (such as Qiagen's miRNeasy kit) may decrease variability in your results. PMIDs for a couple of PubMed publications you may find interesting:27321975, 24885883.
I have gone through literature and found that miRvana is most commonly used ( as suggested by Venkatesh) but TRIzol and Tri reagent are also better especially in stored samples.
I have tried Tri reagent and kit and I agree with Venkatesh. I prefer to use a kit (mirVana is very good) rather then Tri reagents because they may lead to miRNA loss. This can be harmfull to profiling and microarray studies. Besides that the quality of the miRNA is higher using the kit.
I also prefer column-based kits than TRIzol. If you are worried about the yield, you can find a slightly modified column-based extraction from McAlexander et al. 2013 and Burgos et al. 2013. One with the addition of glycogen as a carrier and the other basically with an extra re-extraction step, both worked for me
