Hello everyone,
I tried many times the T24 cell line using 2 different transfection agents(Lipofectamine 3000 and Xtreme Gene HP DNA) and using different concentrations of transfection reagents but to no avail. Is there anyone who had been successful in transfecting this cell line?

Concerning hard-to-transfect and primary cells, I would definitively recommend trying an alternative method to "conventional" lipid-based ones. The Magnetofection*** technology should be the method to use.
It is a transfection method based on physical properties using a magnetic field to attract complexes onto the cell surface; membrane integrity is not touched as complexes enter via classic endocytosis pathway.

We propose stand-alone transfection reagents composed of magnetic nanoparticles that can be used for any application (DNA, siRNA, neurons, primary cells, in vivo, virus...) and also a formulation that can be used in combination with any avaliable transfection reagent.

Dear Hamadi,
We use an electroporation system from Bio-Rad, which works very well (with very high efficiency) in our cell lines, but we never tried T24 cells. Maybe you can check if a group at your university owns an electroporation system.

Fu WJ et al , used calcium phosphate-DNA co-precipitation method to transfecting fusion gene into T24 cells .
