In my lab cells are cultured in DMEM and for certain experiments like to see the intake by cells we use opti-mem media. But my new cell type is being cultured in RPMI-1640 with 1mM sodium pyruvate and 2mM L glutamine. Can I use opti -mem for this cell also which is cultured in RPMI for intake studies?
I agree, it could not be harmful to replace DMEM by OptiMEM for a short time.

If you will made sudden transition, cells may go under stress or senescence. I would suggest to slowly acclimate cells to Opti-MEM using different ratio with RPMI and increasing Opti-MEM conc. at every change untill you arrive on pure Opti-MEM. You can also thaw a new stock of your cells in Opti-MEM and hopefully it will grow fine.

Yes surely you can use. Opti-MEM is a serum reduced media and not serum free. So we often use it during Lipofection experiments but the point is you cannot keep your cells in Opti-MEM for longer duration. You have to optimize it and preferably 4-6hours works fine.
