I want to prepare human membrane proteins. Expressing the protein in yeast, insect cell or in cell free extract which would give me the best result?

Among the three which would give the best result with highest yield and why. Considering post translational modification and glycosylation is necessary for our membrane protein.

GMO are one possibility, but if you consider insect cells, have you also considered human cell lines, either "as delivered" or overexpressing? Costs (material and personel) of both methods can be comparable, depending on expression level achievable.
