I am trying to make an LNP-mRNA system using a modified protocol from Semple, SC et al. "Rational design of cationic lipids for siRNA delivery." My specific question entails the method of siRNA solubilization. The protocol asks for siRNA to be solubilized in 50mM citrate, pH4/ aqueous solution containing 30% ethanol. My concern is that I will be using mRNA not siRNA and a pH4 would severely degrade my mRNA as it is single stranded. Thoughts or advice?
