i want to put MTS on suspended cells to evaluate viability but the data sheet demonstrate the details for attached cells but not demonstrate the fine detalis for suspended cells and there is some problems meat me during experiment for example after culturing cells for 2 days i found that some wells media had evaporated and others decreased.
Hi Aya,
When you say 'evaporated' , do you mean the wells dried out completely? This should not happen but if this was the case, check the water tray of your incubator, there's probably no water left! Also, to avoid evaporation almost completely, especially over the course of a few days, use only the middle wells of the plate for your samples and fill the surrounding wells with PBS or media. This way you should have minimal variation in the volume of your wells after 2 days.
In your concrete case, what you would need to do is to adjust the volume of MTS based on the volume of your media, e.g. if you normally add 20 ul of MTS to 200 ul media, you now need to add 10 ul to 100 ul.
