Even if the status of p53 is known in the cell line, one may require to optimize the concentration of hydrogen peroxide for a given time. Additionally, culture conditions (medium and growth factors) may influence the outcome.

Use different doses of Hydrogen peroxide starting from 100 micromolar to 1 mM, In my opinion you can use 100-500 and 1000 micromolar concentration, see which one gives best result. Can also do a time kinetics with only one dose 3,6,12,24 h.
