I am culturing one kind of cells with some stem cells criteria.
While I was preparing embryoid bodies in Petri dishes from these cells, they were attached to the bottom and grow naturally and now 80% confluence.
What are the approaches to study the extracellular matrix properties of these cells? that enabled these cells from this amazing action.
I believe fibronectin is a big one. I am working on PBMCs and I am interested in monocyte derived macrophage. only monocyte is able to adhere to plastic surface but not the other mononuclear cells. the main cause most likely to be fibronectin. To examine that try to use a model of these cell with a knocked-down/out fibronectin gene and allow them to attach again and see.
you are the most welcome!!. Also you can apply this knockdown/out system using siRNA or CRISPR to examine anything (genetic based). also you can track the cytoskeleton and ECM by in-situ hybridization technique (fixed cells) or you can use tags and specialized microscopy for living cells.
