Does anybody know that how I can induce immune suppression in balb/c mice using drugs or any other simple method? Needless to say I cannot use nude or SCID mice in our conventional animal lab. On the other hand, I want to establish cancerous mice using human cancer injection after I get immunosuppressive balb/c mice. Thanks in advance.

I have worked on inducing the immunosupression in Balb /C mice.
Take cyclophosphamide and cyclosporine to induce the supression in
the doses discribed below:=
To asses the B lymphocytes you should take 250 mg/kg p.o. single dose 2 days before the immunization in the mice.
To assess the T lymphocyte take 5 mg /kg cyclosporine p.o.x 5days after the immunization.
According the protocol take the titre and read the reading.

You can try B cell, T cell and NK cell depletion using anti-B cell, anti-T cell and anti-NK cell antibodies.

Hello all! We are working on efficacy experiments for chronic T.cruzi infections. To detect if parasites are still there after treatment, we have been immunosuppressing with cyclophosphamide, either 2-3 doses, spaced about 48H. This seems to work great for allowing the parasites to come back (we can detect them via IVIS imaging of a luciferase gene about 20 days after the first dose of cyclo.) The problem is that this appears to leave them open to either an oppertunistic infection or some other complications that is leading to urinary problems, prolapsed vaginas, and death in >30% of the treated mice.
We have tried dose titrations, but it seems we need at least 150-200 mg/kg cyclophosphamide to get the parasites back to detectable levels, and this is where we pick up other complications. Any ideas how to avoid these undesired side effects, or is there an alternative to cyclo we could try which might be more tolerated? Many other groups around the world use this cyclophosphamide regimen without issues, so I am worried we are dealing with some latent opportunistic infection in our facility.....
