A few points: Usually 1-5x10^6 breast cancer cells are typically injected either in the mammary fat pad or subcutaneously in immunocompremised mice (nude, athymic, NOD/SCID, NSG, but not ICR!). Once a tumor forms, you will see a bump growing and since these mice don't develop spontaneous tumors subcutaneously or in the fat pad, this is for sure your cancer.
1 x 107 is a very high number of cells for tumor formation. As Richard mentioned, 1-5 x 106 cells are generally sufficient for formation of tumors either subcutaneously or into the mammary fat pads of either NUDE or NOD/SCID mice only. Subcutaneous or mammary fat pad tumors can be easily detected as they start forming a bulge in 10-15 days as they grow.
If you plan to inject SKBr3 cells intravenously into the lungs, you can implant upto 1.5 x 106 cells at the maximum. Tumor formation in this case can be predicted by the weight loss of mice.

SK-BR-3 cell line is a human breast adenocarcinoma. You can't use Balb/c mice for inoculation. You should use Balb/c nude mice or B17.Scid or NOG scid or NSG scid mice or ICR scid mice.
