Im doing ELISAs with a number of samples and then I have my results as ODs and AUs. When i repeat the exactly same ELISA with the same samples, i see the ODs change. My samples come from patient sera.
You are not indicating what are you measuring with your ELISA test neither if you are repeating it in different days, how are you storing the samples... so many factors could be affecting your measures here. Do the internal controls have the same values? ELISA ODs can change from one experiment to another, that's why you always include a subset of controls and refer experimental values to them.
Hello Roser, you are right. Im measuring antibodies and the internal controls have the same values. My samples come from -80ºC and the first experiment was done a year ago.
