The DiO staining depends on your purpose. If you want to visualize mitochondria in live cells, obviously your cells are useless. Rhodamine123 has a similar property to DiO for the cell viability in many cell types. In particular, sperm cells were stained with these dyes for viability evaluation as a vital staining.
However, if your aim is to visualize the endomembrane system within the cells at higher concentration, the fixed cells would be OK for that purpose, visualizing ER, probably Golgi, and other membrane vesicles.
