I would like to inject 250 ug of BFA i.v. into a mouse and at the dilution step from neat DMSO to 10% DMSO in PBS the BFA crashes out of solution (BFA is at 1.25 mg/ml at this point). Usually this protocol is employed for tetramer/cytokine staining. Any suggestions?

Dear Anthony, BFA is very poorly soluble in water; maximum solubility in plain water is about 50-100 µM. Therefore, maximum concentration in water can not be more than 28 mkg/ml. So, you will need to inject suspension of BFA i.p. Another way is to dissolve BFA in DMF and, then, in PBS. In DMF:PBS mixture (1:1) BFA has solubility of approximately 500 mkg/ml. However, I have some doubts to use such mixture for i.v. injections.
