We are planning to purchase a DNA Synthesizer, and we are evaluating two equipments, the Coliseum 96 DNA RNA Synthesizer (Geneforge incorporated company) and the ABI3948 which is only 48 columns but already includes purification. Has anyone any reference or opinion for any or both equipments? Is it easy to get spare parts? Low reagent consumption? We will use the equipment for RNA / DNA oligos, modified, Taqman probes, and long oligos for gene construction.
Probably the world expert on DNA/RNA synthesizers is Mike Gait at the LMB Laboratory of Molecular Biology, Cambridge, UK.

Ask for a demo unit (for both if possible) before purchasing. Then you will really know how long of an oligo they can make accurately (analyze by MALDI), and you can answer all your other questions too.
Thanks Gary, Unfortunately is not possible to have a demo unit since we are located in Mexico and the equipments in USA.
Then I would ask them for a list of Schools/companies (with contact information) that they have sold them to and call them up to see what they think. I would also compare their reagent's cost and see if you "have to" buy reagents from the manufacturer, or if you can get them from other suppliers (usually much cheaper). ABI has been around a long time so there should not be any issues with their equipment. If I was making synthetic genes, the maximum length that I could make an oligo would be very important. Off-line purification should just be a spin column.

The other thing I would do is figure out the cost/oligo for each machine versus buying them from Operon (one of the best, that also checks every oligo by MALDI to confirm the correct product was made-very important). Operon can also make modified oligos. I was once in a lab that bought an oligo synthesizer, only to have the cost of commercial oligos drop making the machine more expensive when you figured in reagents and operation costs.

- based on my knowledge the Company Geneforge is out of the market since end of 2012
- the abi 3948 is available as a used instrument, but you will have problems with consumables and spare parts, because the original producer of this instrument(Applied Bio Systems) is out of the dna synthesizer market for may years
- I assume you are interested in the abi 3900
- the abi 3900 is a very reliable instrument for dna-synthesis
- my experience with the abi 3900 (as an oligo producer and as service provider for dna synthesizers: I can fully recommend the abi 3900
