We have an approach that we are evaluating in a mouse model of stroke which stimulates vessel development. We would like to show that vessel development is the key mechanism responsible for the functional improvements that we see after the delivery of our intervention. I recognize that there are many VEGF antibodies are available but I don't want to have to deal with the challenges of requesting commercially restricted agents from a drug company. I would much prefer to purchase something from Sigma or Tocris. Any suggestions for effective and affordable candidates?
MMPs are required for new blood vessel development. Maybe there's a good dual MMP2/MMP9 inhibitor out there. Haven't tried though.
It's still gonna be more expensive than a small molecule drug, but a lot of companies offer Abs for on vivo functional use. Specifically i seem to remember Biolegend having something they called LEAF which stood for low endotoxin, azide free products. I'd check with them and maybe abcam/rnd/thermo and see if they may have VEGF abs that would suit you!

The only things I can think of right now are RGD containing peptides. They inhibit av containing integrins (with b3, b5, etc).
Another one is PAI-1 (from Molecular Innovations) that inhibits integrin binding to the vitronectin RGD sequence.
Any compound that inhibits platelet activation could also be a candidate.
If you believe that your process is VEGF-mediated (not all angiogenesis is), then there are several small molecule VEGFR2 inhibitors that are both used clinically and available commercially (sunitinib, axitinib, sorafanib, pazopanib, etc.). You might also look into DC101, a neutralizing anti-VEGFR2 antibody that works in mouse. Finally, adenovirally-delivered soluble flt1 is also effective. The affordability of each of these depends on your lab's skill set and definition of affordable.
