Interested in depleting microglia using PLX3397. I have read dozen of studies that formulate this into rat chow - or use oral gavage. I would preferably deliver the drug chronically using a minipump i.p. The drug is soluble in DMSO with PBS. Is there any reason why I should not be able to do this?
Based on my understanding, 3379 should work via minipump. I've used it in vitro and in vivo which suggests that first pass metabolism isn't necessary. In fact, one might be able to limit peripheral exposure to some degree, thus preserving FLT3 and Ckit expressing in marrow, for instance, as well as many csf-1 expressing monocytic cells.
I have heard back a variety of mixed responses for obtaining access to the drug from Plexxicon but understand that it can also be sourced from a variety of other suppliers. I would love to know what other peoples experiences have been?
