There are lots of factors can influence the Ct value:

1 How about the RNA concentration you use for the reserve transcription?

2 Is there any genomic DNA contamination in the RNA sample?

3 Are all the q-PCR use the same cDNA?

It might help:

It was around 1500-2500ng/ul. Fixed at 2000ng/ul as I did in my earlier experiments.
