I'm currently working with estrogenic compounds, and the manufacturer recommends to elute the 17β-Estradiol powder in this way add 1 ml absolute ethanol; gently swirl to dissolve; add 49 ml sterile medium while mixing.
But I would like to know if I can elute it in 50 mL of absolute ethanol, and prepare different long term storable work solutions.
I am guessing that you wish to work with cell cultures. The manufacturer recommendation is probably in order to prepare a stock solution with a relatively low concentration such as a 20 μg/ml stock solution (recommended by Merck). I guess that the addition of the sterile medium is to make sure that when preparing the working standards the ethanol percentage is sufficiently low.
On the other hand, 17β-estradiol has a solubility of 2.5 mg/mL. So you can probably start with 1 mg/mL in 100% ethanol, filter, and then further dilute, enough so that the final concentration of ethanol is less than 0.05%.
