I wonder which basal medium is more efficient in inducing osteogenesis of mouse MSCs. High-glucose without FBS supplement is recommended by my lab, yet other reports suggest that low-glucose DMEM + 10% FBS is the standard basal medium for osteogenesis.
There's a related report showing a higher tracer uptake in low glucose DMEM than in high glucose DMEM [1]. Can this report help me decide which is better regarding osteogenic induction?

Normally for osteoblastic differentiation we use DMEM or commercial media with low glucose..because high glucose content in media will lead to chondrocytic lineage..it is proved already..so its better to use low glucose..
We use a commercially available osteoblast differentiation medium. It contains 3.15 g/L of D-glucose, which is closer to high glucose DMEM. It is also supplemented with FBS.

Our Lab uses alpha mem essenitals medium ( minimum essentials medium eagle from sigma) for differentiating our mouse MSC to osteoblast. The specs do not specify if glucose is present, however we always get good osteogenic differentiation. We also use hypoxic conditions as well.

By the way, MEM from sigma has 1 g/L of glucose.
We use low glucose DMEM (From Life Technologies) for our application and it works well. You may refer to one of our publication for more information.

in general, regardless the type of media you use, there will be osteogenic differentiation, however, some may enhance this process, low glucose media with addition of other factors such as dexamethasone, beta-glycerol phosphate and ascorbic acid, seems to be the best...
