I'm working on senescence and I'm inducing it using TNF alpha. I would like to assess the induction of the phenotype looking for the cytokines typical of the secretome. Which cytokines do you suggest to start with?
There is no specific cytokine that can be used as a marker for senescence. Combination of one or more features like Cell-cycle arrest (increase p16, and p21 levels), resistance to apoptosis, SASP, flat morphology (for in vitro study), increased Beta-galactosidase activity, and DNA-damage markers (H2AX) can be used to monitor senescence. https://pubmed.ncbi.nlm.nih.gov/32961620/
Different stresses induce differential senescence markers depending on the tissues or cell types. So not all markers can be seen in every cell type. most of the time we have to wait for a long time to induce senescence (10 days to ~2 weeks for in vitro study). Hope this answer is helpful to you.
