I have to analyze Eruca sativa roots and leaves. I've seen some methodologies that involve the utilization of the SPI-Dry Critical Point Dryer, however, I don't have access to it. Can I use a Dry Box as an alternative and obtain a similar result?
Drying depends on the detail in which you are interested in the SEM micrograph. Please let me know , what exactly you need to analyse in SEM scans
As I work with chitosan-based nanofertilizers, I'm trying to see the nanoparticles in the roots.
Hello,
You can use HMDS (hexamethyldisilazane). After 100% ethanol dehydration, replace ethanol by several baths of HMDS then you let it evaporate.
Work under fume hood!
I would second what Nicolas Barois mentioned above.
Just a warning: HMDS is a bad stuff, volatile. Please use gloves and work under fume hood. Also read the manufacturer's guide lines. Good luck!
Thank you for the advice! I'll work carefully with the HMDS. Chandrasekaran Nagaswami
