Good evening, I am working on determining the karyotype of a beetle species and performing an in situ hybridization. I have difficulty obtaining good chromosome spread (especially in the summer months and I don't understand why). I work on embryos, with colchicine and sodium citrate for a hypotonic solution. Could anyone give me some suggestions? I would be deeply grateful..
