Hello, my work involves growing cells in the matrigel and for recovering cells from matrigel i usually use cell recovery solution from corning. As now i am curious to know if theres an alternate to recover the cells from matrigel. The mechnical pipetting doesnt work well for me. Please do suggest if theres an alternate methodbt

which cells are you using? 3D organoid cultures? I usually use a very short 2-5 min) incubation with TrypLE (Gibco, prewarmed) at 37C (not too long to avoid effective dissociation of your cells! So optimize accordingly), as the proteases dissolve Matrigel easily. Another method is using ice-cold (
Bart Hee, Thank you for you suggestion. My work involves Retinoblastoma cell line,Ncc rbC51. I tried repeated pipetting with ice cold EDTA-PBS and the cells seems to get pelleted along with the matrigel.
Bart hee, I have tried to recover the cells from matrigel using ice cold Cell culture media and it WORKED so well. Thank you for your valuable suggestion. :)
