I am observing a large necrotic population in glioblastoma cells that I am culturing in 10% DMEM medium. In spite of changing all parameters of culturing and making sure of the sterilitu, the cells look as though bursting and give out black debris like structures. Any reasons or solution to this?

Glioblastoma cell necrosis is characteristic of a tumor obtained from an organism (animal or human) when the cells adapt to the conditions of the culture environment. Glioblastomas from patients are a genetic tumor made up of different cell populations, especially if the cells are obtained from the center of the tumor. In the central part of the tumor, necrosis is observed as a result of impaired transport of oxygen and nutrients due to diffusion and against the background of rapid proliferation of tumor cells along the periphery of the tumor.
For example, for human glioma U251, no necrosis is observed and the cells grow well in culture.

Whatever Dr.Alexandr has mentioned is right. When you need patient tumor for cell culture avoid taking the slice from the center of the tumor. The central part of the tumor is necrotic. Also avoid fatty and calcified tissue.
