As few study shown RNU6 B in tissue,while few claimed about miR-16 etc....As I have to perform in all the above samples in serum, whole blood and Tisssue samples.Experts comment needed who are currently working on the same.
In my opinion that is something you will need to access with your samples. I assume you want to compare healthy donors with some sort of disease. I have used RNU6B for stimulated culture cells and it worked but miRNAs level of expression are very tissue specific and it can be influenced by different pathologies and also vary from individual to individual.
I have seen an article that says that RNU6B is not a good normalizer for breast tissue and they suggest miR16. There are custom ordered pre prepared plates with different miRNAs. It could be a could alternative to you.
Yes @Andre Fernandes thanks for the suggestion yes I have also seen article on RNU6B few supports while few rejects it in Breast cancer and hepatic tissue.Further few suggest to go with mir-152 and mir-23b ,mir 103, mir 16 etc.But I think it would be better as you suggest, to test few suggested in literature in healthy controls then go for the rest.
I am sure you know, but I would just like to remind that tissue and serum and/or plasma samples miRNA signatures may differ. Andre is right in suggesting you to compare healthy and patient samples. However, I would also like to add that if you cannot procure tissue samples (in statistically significant numbers), I would recommend use non-invasive samples.
I would also like to bring to your notice that interestingly, at times foreign miRNAs can also be detected in circulation.
