I have currently used 4 time periods for looking at my cells for the NF-kappab activation: 5,10,15 and 20 minutes. Looking at the western blot during these specific time points, the increase in the nfkappab p50 subunit is visualized in the cytoplasmic extract of the cells. However p105 is seen, also elevating with the increasing time points in the nuclear part. However there is no p50 in the nucleus. moreover after 24 hours p105 is just present in the nucleus. Cn anyone suggest me what time points i should use for the analysis of such a translocation, and avoid the delay of proper results? Thank you :)
