I am interested in isolating phages from environmental samples and different papers suggest different methods to do so. Many people use SM Buffer and phage buffer to re-suspend the isolated virus particles. Are these universal buffers for phage? Are there other types?

I notice the components of the buffers include salts such as MgSO4, CaCl2 and NaCl2. I know that CaCl2 is required for adsorption for P1 phage. What about the other salts, are they also required for adsorption for other types of phages? Are there other types of salts that are not present in SM/phage buffer that could be essential for phage isolation too?

Should these salts be added in the agar/broth as well?
