Hi,
During biopanning rounds of phage display, after infecting TG1 cells with eluted phages and placing them on 2xTYGA I get inconsistent TG1 colony morphology.
Some rounds the colonies are small and discrete, and some rounds I get large colonies with undefined edges.
2xTYGA media is made fresh in the week before infection and plating, and the transformation efficiency is the same between the rounds, regardless of TG1 colony morphology.
TG1 are grown from a single colony grown on M9 minimal media.
Has anyone encountered this?
