It is possible to examine the conventional sections for light microscopy or ultrathin sections for TEM by SEM after coating (5-10 nm gold, etc). In BSD mode you will have very informative images.
You can examine the sections without coating in VP mode, but it does not have any advantages.
Prakash i am asking for the method to fix the cells for better viewing the cells in SEM. I did the gold coating but the morphology of the cells were distorted when i saw the slides in Fe-SEM.
before anyone will perhaps reply in a polite and serious manner you certainly - with regard to the method (s)/ steps of processing you made until observing in the SEM first (ee your Re #3) - should put your cards on the table...please detailed information then you'll get for sure answers (and: usually one will know how to fix cells for SEM -observation ...)

The method which i followed is: Fix the cells with 3% glutaraldehyde followed by successive washing with 30%, 50%, 70%, 80%, 90% and 95% alcohol. followed by overnight drying at room temperature. But when i am viewing them in SEM the next day, the cells are distorted. I am not able to view the correct morphology of the cells.
