In this image of PFGE, ladder (Lane-1) is decent, but band patterns of the test sample (Lane-2) is neither sharp nor clear. Just need to know how to make it good as the protocol which was followed also used earlier with good results.
Hello Mr Nayak. There are a lot of possible technical reasons. You can kindly refer to the PPT file as attached. It is possible that your mapper was contaminated, or your plug was not prepared as good as your previous experiment. Maybe your plug was stored too long, or contaminated/damaged during storage. It is also possible that the gel imager was not focused during image processing.
Are these plugs were prepared together? if not then as Nurul mentioned there may be quality difference in plug preparation and RD also chances of contamination.
