I want to amplify a 4.5kb fragment from a plasmid. Can pfu polymerase from genebio is suitable for the job?
yes it should be ok. Just make sure to use an appropriate extension time for your amplicon.
It should be fine. You need to test elongation time and optimal temperature. For PFU I usually use 1 min elongation for a 1 kb fragment.
I think it's borderline. You will be more likely successful with Phusion.
