I'm using the millipore TEER electrode to measure the resistance of colon cancer cell line.
I have been unable to get a stable reading, most of the time it shows fluctuations.
Although the transwell looks confluent under the microscope the TEER reading is still around 200 only.
TER depends mainly on cell line characteristics and to a less extent on colture medium. In my hands, dog MDCK may give TER up to 10K Ohm in high glucose medium, while human HEK293 never reach more than 40-50 Ohm. Moreover, the highest reading is obtained just after confluence or few hours after medium change. Later, TER tend to decrease. In my experience, 200 Ohm (clean from blank value) is an accebtable resistance. If your cell are easy to trypsinize, even when confluent, you cannot hope to have a strong TER.
Thanks Lucia, but my senior who worked with the same cell line 2years, he used to get a TEER of 450 ohm.
most of the studies on the same cell line have shown that the TEER comes to 450 ohm which conforms it is confluence.
Regarding your first question, TEER value gives you an excellent idea of your cells differentiation degree, and not all cell lines are able to differentiate in vitro. As Lucia pointed out, there are different factors that could influence TEER values. Caco-2 cells for example, grown in low glucose medium on collagen-coated transwells can reach more than 1000 ohm at day 7 of culture, increasing at day 14 and decreasing a little bit at day 21. TEER should be also measured before changing the medium, so 24-48h days of culture is ideal to measure it prior to change the medium. Another important issue is the type of electrode and the type of the insert (transwell) you use (polycarbonate, etc). For Millipore I recommend you STX100 electrodes to have more reproducible measures. Even im using the same caco-2 cells with low glucose medium but i have the problem of cells coming out , Transwell is now growing for more than 50 days but still the TEER is not coming to 450 ohms.
I dnt knw what is the prblm , im using the Millicell-ERS only for measuring TEER, most of the time when i dip the electrode in the chambers it will be fluctuating, and its even decreasing.
Can you please help me to sort this out
Even if you are not using the electrode, you must take it into a saline solution with electrical characteristic similar to the colture medium (PBS is OK), to have it equilibrated and stable.
Moreover, are you sure you did not change anything, ie cell supplier, FCS batch, so on ?
Its an old question and maybe a stupid answer but did you use your millipore TEER electrode with the battery still in charge ?
Anybody have measure TEER for human brain microvascular endothelial cells before? and this is just a monolayer cell without co culturing with other constituents of BBB. Thank you for feedback.
I did this for heart vascular endothelial cells (MCEC (Cedarlane/CELLutions Biosystems Inc. ; Ref : CLU510) and as they didn't bind with tight jonction, the TEER was not elevated even when arrived at confluence.
