Sp2/0 is a suspension cell line. It is quite robust and easy to maintain. I used to grow it up in a DMEM+FBS 10%, 1%p/s medium. I strongly recommend checking your cells for a mycoplasma contamination since it might affect cells growth.
you can maintain the Cultures in DMEM+10%FBS+PS. You can add fresh medium every 48 hours to the culture flask till it gets confluent. Maintain cell density between 5 X 104 and 5 X 105 viable cells/mL. Some cells can attach and be transferred by shaking them loose. BD T 25 flask will work.
I dont know why?when I seed the cells in flask after defreezing they are alive and they have spherical shape but after a few days, the shape of them will become rectangular???
I'm also working on sp2/0 cells and I had never face such problem. Tell me your preservation method because in my lab revival rate of correctly freezed sp2/0 cells is about 70-80%.
