Whole-blood samples were collected from human subjects with the aim of extracting circulating RNA (including microRNA) from them for transcriptional profiling. My first question is: does any one know whether it is feasible to safely store the whole blood samples in RNase-free environment for a few days without affecting the quality and integrity of the RNA to be isolated? Tissues would be dropped immediately in an RNAlater protection reagent and stored the fridge. According to previous readings, however, people advise to quickly (Within 4 hrs) process the entire blood samples to isolate the plasma, which can then be stored at temperatures such as -80°C. My second question is: would the freezing and thawing processes affect the quality of the plasma if it had been stored without any protection?
