I would like to know What is the best protocol for competent cell preparation?
Any suggestion in this regard would be appreciated.

Hi,
You can follow Inoue method (https://bio-protocol.org/bio101/e143)for preparation of ultracompetent cells and it gives very high transformation effeciency.

We follow this attached protocol. It works perfectly well even for co-transformation.
Rubidium Cl protocol seems to work much better than CaCl2. https://openwetware.org/wiki/RbCl_competent_cell
