I incubated my organism (thermophillic bacteria) on LB broth at 37 and 40 degrees centigrade for the purpose of extracting DNA from the organism but did not show any growth after 22hrs of incubation.I want to try higher temperature like 50 degree for the cultivation.Iam not sure if growing the organism at higher temperature is good for the purpose of extraction of the genomic DNA.
