I'm currently working on a topic related to studying the anaerobic biodegradation of TPH in marine sediments. In this work, sediment should be mixed with fuel oil to a concentration of 20 g (oil)/kg sediment. I am not sure whether to make this on a dry or wet basis. normally the sediment has some water content and it can not be dried because drying will kill these anaerobic bacteria. so if I mix the fuel with the sediment (wet as it is) is that correct?

Once you are using the marine sediments, you can measure the moisture content by fraction of mass loss. use following USGS reference.

Then you can include moisture correction factor for the samples.
But for the comparison studies with the different inoculum or strains, you need to manage the anaerobic condition with artificial incubation environment.
The biodegradation can alter due to several factors such as nutrients, moisture, bioavailability of TPHs (sorption to particles), temperature, oxygen, and presence of TPH-degrading microorganisms.
Therefore, you need to control the environment for performance identification. Then only we can manage uncertainty throughout the experiment.
