Spheroids or spheres derived from cancer cell lines usually consist of proliferating & un differentiated cells. Assuming this, what can be the alkaline phosphatase activity of cancer derived stem cells (in spheroids)? Will it differ in serum free media from 10-20% FBS? How do you measure growth rate of spheroids? I tried but could not dispense the exact number of spheroids in with serum & without serum media to check change in morphology and growth.

So there are a lot of metabolic changes in the spheres as compared to the adherent cells. What I found through proteomic analysis (although I did not perform any enzymatic assays) is that mitochondrial enzymes, and protein involved in translation was is significantly reduced (Kanojia et al., 2012 Proteomics). Second peripheral explaination is that so called "cancer stem cells" are metabolically different than the non-stem cells. For example CSC have higher ability to resist oxidative stress (Diehn et al Nature 2009). Hence in spheres I would expect a difference of metabolism and likely a change in alkaline phosphatase activity. The third question is how do you measure the growth rate of spheroids. You can do MTT (Wan Hong Ho et al., 2012 PLOS one).
