Actually to establish a cell line from cancerous tissue primary culture, the splitting ratio sometimes depend on the no. of cells live in primary culture. To establish a cell line the no. of doubling time required is a primary question to be solved, usually 80-90 doublings are required. But to achieve this, is it necessary to split from 1st subculture only?

You can try from the first and second subcultures (in separate culture flasks) and compare results. You will get different subclones from the original cancer. Each subclone may develop into a cell line. But, Its very possible that second subculture will give rise to clones that may not truly represent the oprimary cancer.
