What is an appropriateness of studying secretome in early passage cancer cells derived from tumor tissue?

I am not sure whether my thinking is in the right way or not but I think early passages of cancer cells are more suitable tumor model of tumors in vivo because after some passages (5-15) cells are altering their genomic and transcriptomic profiles so we may not get those biomarkers (which might be expressed in serum or body fluids in early cancerous stages), which are expressed in early passages if we study the later passages of established cell lines for secretome study.
