The best storage method varies for different members of the Ascomycota. For a number of ascomycetes, particularly the faster growing ones, dried filter paper is a good and inexpensive method (see Peever and Milgroom. 1992. Phytopathology. 82:821-828.). Most fungi we have tried do well with the agar strip method. We have seen loss of virulence with plant pathogenic fungi stored in sterile soil and various morphological changes have been reported by others (e.g. http://128.104.77.228/documnts/pdf2004/fpl_2004_nakasone001.pdf).
Store your cultures in sterile distilled water vials (2-3 ml). Try to transfer only mycelia and if available conidia. Do this in aseptic conditions, carefully. I observed that these tubes can be stored easily for two years in household refrigerator (4-10°C, Not freezer). This is a very cheap and efficient method.
