I had studied that internal transcribed spacer (ITS), Isolation of DNA and PCR amplification and DNA sequencing and BLAST and phylogenetic analysis are used. Can anyone suggest articles about other methods and above methods?
There is a barcode for fungi avalible. For identification morphology is the main however, if you are interested in identification and phylogenetic analysis you have to use more gene sequences like Elongation factor ef1, beta tublin, GAD etc for robustness.
Multilocus genealogical concordance phylogenetic species recognition (GCPSR) scheme can be employed for the accurate speciation of fungi. RPB2, EF-1 alpha, beta tubulin, CAM loci can be assessed for this MLSt analysis.You can refer the paper of O'Donnell 2009 for MLST.
It is depend on your goals. If you need to identify plenty of fungi from the sample, you could use ITS and D1D2 regions. But for species identification in some particular groups you should use additional region, like beta tubulin intron.
Also protein mass-spectrometry MALDI-TOF is a perspective modern method for fungi identification.
