I am generating dendritic cells from mouse bone marrow which I am getting >80% CD11c positive cells on day 8. But using GM-CSF alone for DC generation give CD11c positive cells which are also CD115 positive, interestingly using GM-CSF+IL-4 condition for DC generation drastically reduces CD115 marker (macrophage precursor marker) on CD11c positive cells. what could be explanation for this and what are the other markers that can distinguish immature dendritic cells from macrophages, so that I can be sure that the CD11c positive cells are DC? will DC205 and DC209 work?

Thank you Dr. Julie for the link..it helped a lot..the article has given lot of insights on features of dendritic cells generated from bone marrow precursors with the help of GM-CSF and GM-CSF+IL-4..thanks again, but unfortunately they haven't mentioned about DEC205 and DEC 209 markers on these cells as I am getting high DEC 209 & DEC205 expression on GM-CSF cells but low DEC 209 on GM-CSF+IL-4 cells..
