I want to perform the dual luc. assay using Hl-60 cell line which is a floating cell. Therefore, I explored different papers but no brief protocol found about the assay using this floating cell.
If anyone experienced or idea about this please share.

I have never used the assay with a floating cell line, but I would suggest to centrifuge gently the cells, do a wash with PBS, and after the 2nd centrifugation resuspend in the lysis buffer and continue with the classical protocol. In your case, it's easier to do the assay in tubes rather in a 96-w plate, I would expect.
