Dear All, I would like to check the cell viability of THP-1 derived macrophages after treatment with different stimulants. I am looking for a specific gene that is preferably upregulated in these cells when they lose their viability.
It needs a careful and precise look into the related published studies. Maybe you find studying genes involved in apoptosis and other types of cell death useful. Caspases, cyt-c, Bax/Bcl-2 ratio, etc.
That depends on what kind of death is happening after each treatment. For apoptosis you should blot Caspase 3 as is is a convergent point for both extrinsic and intrinsic pathways. For necroptosis blot phospho-MLKL. For autophagy, LC3-II protein will be elevated. There is no single gene to be checked for.
