Sample was ethanol fermentation. Streaked on 3 plates and place in candle jar for 3 days then at 4 degrees for 4 more days. (saccharomyces cerevisiae vs lactic acid bacteria)
1) SDA = yeast and bacteria
2) MRS + cyclohex = bacteria growth
3) SDA + gentamicin + chloramphenicol = NO GROWTH
I expected yeast to grow on this plate. All three were plated at the same time from the same sample. But no yeast on plate #3. All I can think of is that somehow the yeast need the lactic acid bacteria in order to grow. Any ideas or explanation would be appreciated!
Did you have to incubate all three plates the same way? Saccharomyces cerevisiae is a facultative anaeroble while lactic acid bacteria are aerotolerant anaerobes. The yeast will grow readily in the ambient conditions of the laboratory without recourse to candle jar, but the lactic acid bacterium will not. Secondly, the yeast is not a psychrophile/psychrotroph, why subject it to growth at 4 deg Celsius? I think you should look at these first before drawing the conclusion you are trying to draw.
