Hi all,

I'm trying to validate an ELISA against anti-drug antibodies in which I use a immunized rabbit serum as positive control. Rabbit was a NZR, immunization protocol was "standard" but I don't know the exact protocol, administration was IV.

I'd like to roughly estimate how many percent of total antibodies the ADAs in this serum could stand for, because guidelines recommends a LOD of 100 ng/mL of the specific Ab, but I can't purify serum. So if I can justify that specific ADA stands for at least x% of total antibodies, I can estimate my LOD and justify the sensitivity of my assay.

An immunology specialist told me at least 90% of total antibodies would be ADA after this kind of immunization on heathly NZR- that suits me fine- but he couldn't give me any source of this knowledge. Do you agree with this statement ? And would you have some data or some papers to support it ?
