My alpha-glucosidase enzyme (from SRL) is degrading too quickly when I make working and stock solution. I store it at -4.0 C. How can I prevent it from degrading? Do I need to store it at -20 or -80?
What is the buffer composition in which you are storing the enzyme. You may include sodium azide (0.02-0.05% w/v) in your buffer to prevent the bacterial growth which may degrade your protein. If you want to store the enzyme in -20, dialyze the enzyme solution against the storage buffer containing 50% glycerol to avoid freezing of the solution or add equal amount of pure glycerol to your enzyme solution.
I'm storing it in sodium phosphate buffer of 50mM. But I'm not intending to keep the enzyme solution for a very long time. Just need it to work for 3-4 days. My stock solution is about 80U/ml and I'm working solution is 1U/ml.
